Urinary uracil as a biomarker of dihydropyrimidine dehydrogenase phenotype in cancer patients

Abstract

Biography

Sihan Wang is a PhD candidate in the Department of Molecular Medicine and Pathology at the University of Auckland, New Zealand, supervised by Professor Nuala Helsby. Her research sits at the intersection of pharmacogenomics and clinical pharmacology, addressing the urgent need for comprehensive pre-therapeutic screening in oncology. Building on her Master’s research into urinary biomarkers for 5-fluorouracil toxicity, Sihan’s doctoral work investigates the broader catabolic disposition of fluoropyrimidines. Her research challenges the conventional focus on anabolic-driven toxicity in mitotic tissues, exploring how alternative metabolic pathways may contribute to adverse effects in non-mitotic organs, such as the heart. This is critical, as current management often requires the cessation of treatment when toxicities arise, leaving patients without effective oncological control. By utilising LC-MS methods to characterise the pharmacokinetic behaviour of 5-fluorouracil in clinical cohorts, Sihan aims to identify endogenous biomarkers for metabolic phenotyping. Her work seeks to establish a pre-therapeutic screening strategy to identify individuals at risk of varied fluoropyrimidine toxicities, thereby personalising treatment and enhancing patient safety. Sihan is interested in pursuing a career in clinical research.
